Partial liver transplantation is becoming one of the standard methods for treating patients with end stage liver disease. Regeneration of small parts of the liver plays a key role in long term survival. Study of the effects of immunosuppressants, such as cuclosporin A (CyA) and FK506, on liver regeneration is very important. Male Wistar rats around 200 g were stidied. They were randomly assigned into control, cyclosporin A (CyA) and FK506 (FK) groups. CyA (10 mg/kg/day) or FK506 (0.2 mg/kg/day) was given orally to CyA or FK group rats respectively from 2 days before surgery until the day they were sacrificed. Partial hepatectomies (about a seventy percent hepatectomy) were performed on the experimental groups. Then they were sacrificed at 6, 24, 48 and 72 hours after hepatectomy. We measured (1) white blood aell and absolute lymphocyte counts in the peripheral blood; (2) ornithine decarboxylase (ODC) content in the remant liverl and (3) 5-Bromo-2-Deoxyuridine (BrdU) incorporated hepatocyte counts as the BrdU incorporated index. We found that the ODC contents and BrdU incorporated index increased markedly in the early stage after partial hepatectomy. The administration of CyA or FK506 enhanced the increase of these regeneration indicators significantly (p<0.05). We suggest that immunosuppressants such as CyA and FK506 can increase liver regeneration after partial hepatectomy in rats. |
