RUNX3 Hypermethylation in Remnant Gastric Cancer: A Preliminary Report

RUNX3 (PEBP2alphaC/CBFA3/AML2) is a transcription factor downstream of the TGF β signaling pathway. RUNX3 knocked-out mice showed hyperproliferation and hyperplasia in gastric epithelium. Lack of RUNX3 expression, partly due to promoter hypermethylation, had been associated with the human gastric cancers. This study is aimed to evaluate the relationship between RUNX3 expression and human remnant gastric cancer. The methylation status of RUNX3 promoter in human remnant gastric cancer was investigated by methylation-specific restriction enzyme digestion and PCR amplification. We retrospectively reviewed the surgically removed specimen of eleven cases of remnant gastric cancer. Hypermethylation of RUNX3 in its promoter region was found in the tumor tissues of all eleven cases. Further studies are required to clarify the direct relationship between RUNX3 and remnant gastric cancer.
