篇名 Pretransplant Mixed Lymphocyte Reactivity (MLR) Fails to Predict the Outcome of Islet Allotransplantation in Congenic Mice

摘要 Congenic mice are known to have different responses to islet allo-transplantation. We studied whether the pr叫ransplant primary mixed lymphocyte reactivities (MLRs) would predict outcome. Splenocytes (responders) from 30 hemisplenectomized B10BR (H2-k) recipients were cultured with irradiated stimulators from C57BL/6 (H2-b )。r DBA/2J (H2-d) donors before islets allo-transplantation. The values of pretransplant MLRs showed a diversity within one order of magnitude (stimulation indexes: S.I. =3.0-12.9). However, there was no correlation between MLR responses and outcome of transplant in terms of graft rejection (n = 13), acceptance (n = 6), or primary nonfunction (n = 3). The remaining mice were excluded because unexpected death (n = 3) and poor technical control (n = 5). The inability of pretransplant MLRs to predict the allograft survival can be interpreted by the limited diversity of MLRs among littermates of inbred mice, and by the unsteady dynamics of MLRs following pretransplant manipulations.
